The Serie A table toppers thus dumped Frankfurt 5-0 on aggregate having won the first leg 2-0.
It was Napoli first Champions League quarterfinal.
Osimhen opened scoring in first half stoppage time when he rose high to head home a measured cross from the right.
It was his third goal of the Champions League having also scored in the first leg in Germany.
He doubled Napoli lead in the 53rd minute for his 23rd goal in all competitions this season after he slid home a low cross from the right.
His left wrist will have to be re-examined in the coming days after he hurt it while scoring this goal.
He will enjoy a standing ovation when he was replaced late in the game.
